「情報から制御へ:ゲノム情報収斂型創薬科学」

 当該21COEプログラム、ケモゲノミクス領域、藤井グループでは“ゲノム科学とケミストリーの融合”を計り、ゲノム情報を集約して見いだされる創薬標的に対してケモゲノミクス(ケミカルバイオロジー)を基盤にした創薬化学の教育・研究拠点形成を目指した活動を推進した。
 具体的にはケミカルプロテオミクスに立脚した創薬基盤の革新という一つの座標軸を設定し、1)包括的機能ゲノミクス・バイオインフォーマティクスを活用した創薬標的の化学的同定、2)環状ペプチドをリエゾンとする高分子ペプチド蛋白質の低分子化、3)アルケン型ペプチドイソスターの精密有機合成手法の確立とケミカルプローブとしての応用、4)膜貫通型受容体、トランスポーターの有機化学レベルでの機能解析、5)バイオインフォマティクスを基盤とした新興・再興ウイルス性感染症に対する創薬戦略の開拓、という項目の有機的統合のもとにゲノム情報収斂型創薬研究を推進した(図1)。
(1) 新興・再興ウイルス性感染症に対する膜融合阻害剤の開発研究:
 I-型、膜融合機構を利用するHIV-1に対して独自に開発したXEEXXKKコンセプトを活用して強力且つ特異的な活性を有する多剤耐性克服型膜融合阻害剤T20EKを開発し、さらに同様な戦略を用いて猫エイズの病因ウイルスであるFeline Immunodeficiency Virus(FIV)およびSARS-CoVに対して細胞レベルで抗ウイルス活性を有するFIVC35EK、SR9EK13をそれぞれ開発した。一方SARS-CoV膜融合阻害剤の研究を通じて直接間接経路のみならずエンドサイトシスを介する間接的感染経路の防御の重要性を再認識し、今後の鳥インフルエンザ、エボラ出血熱、成人T-cell白血病、はしか等の他の新興・再興ウイルス性感染症に対する膜融合阻害剤開発研究に対する有用な基礎的知見を得ることができた(図2)。
(2)アルケン型およびフルオロアルケン型ジペプチド等価体の合成と応用:
 アルケン型イソスターおよびフルオロアルケン型イソスターの(Z/E) 立体選択的精密合成手法を開発し、α4β1型Integrin拮抗剤、GPR54作動剤, CXCR4拮抗剤等の各種生理活性ペプチドへ導入してその力量に関する検討を続けている。最近、ペプチドトランスポーター、PETP1の基質認識特性の同定に応用することができたが、本研究を通じて、フルオロアルケン型イソスターのバイオイソスターとしての特性を厳密に精査することの必要性を明らかにした(図3)。
(3)CXCR4受容体特異的拮抗剤の創出と応用3)
 我々が世界に先駆けて見い出したCXCR4-ケモカイン受容体のペプチド性拮抗剤をリードとして特異な二価亜鉛錯体構造を有する非ペプチド性拮抗剤の創出に成功した(図4)。また一連のCXCR4拮抗剤をケミカルプローブとしてCXCR4と生体内生理および病理との関連を精査すべく、とりわけ癌の化学療法への応用に焦点を宛てて研究展開を図っている(図5)。国内外の多くの研究者との共同研究により、CXCR4は膵臓がん、乳癌、メラノーマ、膀胱がん、小細胞肺がん等の固形がんおよび、急性リンパ性白血病、慢性B-細胞白血病等の血液がんにおける転移抑制、増殖抑制のみならず臨床現場で大きな問題となっている骨髄ストローマ細胞依存性多剤耐性(Cell-adhesion Mediated Drug Resistance)を克服できる可能性明らかにした。


おわりに:システムケモセラピーの必要性
 日本の薬学研究は、“くすりの科学と疾病の治療”に軸足をおいた学術研究に貢献することにより人類の健康と福祉に奉仕することをミッションの一つとしてきたと理解している。メタボリックシンドローム、新興・再興ウイルス性感染症、癌、アルツハイマー等々、有効な化学療法が確立されていない疾病は多い。換言すると“くすり”では治りにくい疾病に対する治療薬開発だけが未解決の問題として残っている。疾病とは内因性のものも感染症等による外因性のものも含めてヒトという生命体のシステムとしての破綻であり、これらの難治性疾患の持つ破綻したシステム自体の堅牢性(Robustness)や構成因子(Multi Factors)の相互補完性、冗長性(Redundancy)は分子標的医薬やEvidence-based Medicine(EBM)といった単純な開発コンセプトをはねつけてしまう。例えば癌の転移一つとってみてもそこには数多くのその要因となる創薬標的の存在が指摘されているがこれを制御できていない。すなわち、これらの難治性疾患の多くは多因子疾患(Multifactorial Disease)であり、その克服の為には臨床の場で経験的に行われてきたカクテル療法をバイオインフォマティクス等の科学基盤の基に再構築し、それに裏付けられたシステムケモセラピー(多元的化学療法)の確立に真剣に取り組む時期に来ていると認識している。ゲノム関連科学にはこのような現状のブレークスルーが期待されているが、現実には創薬を錦の御旗に掲げた各種大型研究プロジェクトは必ずしも期待通りの成果をあげていない。この様な状況下に著者が所属する京都大学薬学研究科では2007年4月1日より「医薬創成情報科学専攻」を設置し、生命情報科学と創薬科学の融合を目指して新たにシステムバイオロジー分野およびシステムケモセラピー分野を開設する運びとなった。
